Enzymatic fluorometric methods for the microdetermination of hexose phosphates in muscle.

چکیده
Our current investigations on the chemical processes associated with muscular activity (1) require more sensitive methods for the determination of various metabolic intermediates than are currently available, and the present work is a contribution to the development of such procedures. These aim at the determination of hexose monophosphates on the one hand, fructose diphosphate and triose phosphates on the other. The methodology consists of a modification of the procedures of Slater (2), by the use of fluorometry for the determination of the oxidation of reduced diphosphopyridine nucleotide. To accomplish this with the required reliability, a number of experimental details had to be critically examined.’ As will be recalled, the Slater methods rest upon the following reactions : Enzyme Preparation A:

The Role of Myokinase in Transphosphorylations* I. the Enzymatic Phosphorylation of Hexoses by Adenyl
The enzyme which catalyzes the phosphorylation of glucose and fructose was described in 1927 by Meyerhof (1). He found that muscle extracts which split polysaccharides and hexose phosphates to lactic acid but were unable to ferment hexoses could be enabled to ferment glucose and fructose by addition of a protein fraction from bakers’ yeast. The same protein fraction activated the aerobic oxidat...
متن کاملSemiautomated assay of normal concentrations of urinary glucose by an enzymatic fluorometric technic.
A semiautomated, enzymatic, fluorometric method for the determination of the low glucose concentrations normally present in urine from subjects in the fasting state is described. The method is developed from a manual, enzymatic, fluorometric technic for the determination of urinary glucose using hexokinase (HK) and glucose-6phosphate dehydrogenase (G-6-PDH). The procedure is based on measuremen...
متن کاملFructose 2,6-bisphosphate in rat skeletal muscle during contraction.
Fructose 2,6-bisphosphate and several glycolytic intermediates were measured in two rat muscles, extensor digitorum longus and gastrocnemius, which were electrically stimulated in situ. Both the duration and the frequency of stimulation were varied to obtain different rates of glycolysis. There was no relationship between fructose 2,6-bisphosphate content and the increase in tissue lactate in c...
متن کاملR . Van Heyningen and a . Pirie
Pirie, van Heyningen & Flanders (1955) showed that the concentrations of certain acid-soluble phosphates in the lens, when estimated by the enzymic method of Slater (1953), were much lower than the concentrations reported by Nordmann & Mandel (1952), who used the barium-fractionation method of LePage (1948, 1949). This method has, until recently, been the accepted and standard one for estimatio...

such as the process of cellular respiration, which is scavenged by enzymatic and non-enzymatic antioxidant systems. Intake of antioxidants from food sources has always been considered in nutrition research. Therefore, the aim of the present study was to investigate antioxidant effects of crocin on the expression profile of genes involved in the enzymatic antioxidant systems in muscle cells. Ma...
